Phage Display and mRNA Display

Phage and mRNA Display Technologies for Antibody and Peptide Discovery

Phage display and mRNA display provide complementary approaches for identifying antibodies, antibody fragments, and peptides against diverse target classes. WuXi Biology and Testing combines established phage display capabilities with mRNA display to support library-based screening, hit identification, and downstream validation.

Our phage display capabilities support antibody discovery across Fab, scFv, and VHH formats, as well as peptide discovery using off-the-shelf and customized libraries. For peptide research, mRNA display offers a complementary cell-free approach that provides access to high sequence diversity and diverse macrocyclic library formats. Integrated assays including biophysical, structural, and peptide validation capabilities can further support peptide development from initial hits into lead optimization.

Phage Display

Phage display links displayed peptides or antibody fragments to their encoding DNA, enabling iterative selection and sequence identification from highly diverse libraries. WuXi Biology and Testing supports phage display for both antibody and peptide hit discovery, with flexible library choice and screening strategies that can be adapted to target characteristics and study objectives.

Capabilities include off-the-shelf and customized libraries, protein- and cell-based panning strategies, multi-round enrichment, binder identification, decoding, and downstream hit validation. Identified hits can subsequently progress into affinity characterization and optimization using complementary biology and biophysical capabilities.

Antibody Discovery via Phage Display

Our phage display platform supports antibody hit discovery across multiple fragment formats:

  • Fab (Fragment, Antigen-Binding, ~50 kDa): moderate stability and tissue penetration; applicable to multiple areas including diagnostics, toxin neutralization, and antibody-drug conjugates.
  • scFv (Single-Chain Variable Fragment, ~25 kDa): compact antibody fragments with high tissue penetration; applicable to CAR-T constructs and molecular imaging probes.
  • VHH (Nanobody, ~15 kDa): highly stable single-domain antibody fragments with high tissue penetration; applicable to targeted drug delivery, in vivo imaging, and biomolecular research probes.

The platform supports off-the-shelf libraries as well as customized approaches based on study requirements. For VHH discovery, both naive and immunized library strategies are available.

Our standard workflow includes multiple rounds of library panning, followed by binder identification using ELISA and sequence decoding, antibody protein production, and binding affinity determination using approaches including titration ELISA and surface plasmon resonance (SPR). Following hit identification, affinity maturation and humanization strategies can support antibody development toward optimized antibody candidates.

Peptide Discovery via Phage Display

Our phage display platform also supports de novo discovery of peptide hits using off-the-shelf and customized peptide libraries. Available library formats include:

  • Linear peptide libraries: multiple sequence lengths available for broad peptide sequence exploration.
  • Monocyclic peptide libraries: disulfide-cyclized formats designed to provide increased structural constraint.
  • Customized peptide libraries: tailored library designs can be developed based on target characteristics and discovery objectives.

Protein- and cell-based panning strategies support hit identification across different target types. Following enrichment and sequence decoding, selected peptide hits can progress into resynthesis, binding confirmation, functional testing, and further optimization.

Phage display can also be used alongside mRNA display and peptide DNA-encoded libraries (DEL), providing complementary approaches for peptide hit discovery and subsequent optimization.

mRNA Display

mRNA display is a cell-free in vitro selection technology that covalently links each peptide to its encoding mRNA, enabling screening of highly diverse peptide libraries without the constraint of a cellular display host. The platform supports macrocyclic peptide discovery through flexible library formats:

  • Linear peptide libraries: randomized peptide sequences for broad sequence-space exploration.
  • Monocyclic peptide libraries: peptide scaffolds cyclized through disulfide bonds.
  • Chemically cyclized libraries: peptide scaffolds cyclized using synthetic chemical linkers.
  • Macrocyclic peptides: library formats supporting macrocycles up to 20 amino acids in length.

The workflow combines library generation, peptide translation and mRNA linkage, cyclization, target selection and counter-selection, iterative enrichment, and sequence analysis.

The platform has demonstrated discovery of both orthosteric and allosteric peptide binders. Downstream characterization, including alanine scanning and cysteine-to-serine mutation analysis, can help identify residues important for binding and cyclization and guide subsequent peptide optimization.

Hit Characterization and Optimization

Hits identified through display technologies can progress into downstream characterization and optimization using complementary assay, biophysical, and functional capabilities. Depending on the molecular format and study objectives, capabilities include:

  • Hit production and resynthesis to enable experimental confirmation and further characterization.
  • Binding characterization using complementary approaches including SPR and other biophysical assays.
  • Functional assays to evaluate biological activity and support hit prioritization.
  • Antibody optimization including affinity maturation and humanization strategies for selected antibody hits.
  • Peptide hit validation through parallel peptide resynthesis, LC-MS characterization, ASMS-based binding assessment, and affinity ranking.
  • Peptide optimization supported by integrated synthesis, assay, and structural characterization capabilities.

For peptide discovery studies, Direct-to-Biology workflows can further link display-derived hits to high-throughput synthesis and experimental validation, thereby supporting iterative design–synthesis–test cycles.

Case Study: Hit Rate and Structure-Activity Mapping

A recent internal case study illustrates what the platform can deliver in practice:

  • SPR-based affinity: binding affinity was determined for two peptide hits using SPR
  • Binding modes: the screen identified both orthosteric and allosteric binders
  • Hit rate: reached close to 60% in this case study (defined as KD < 20 μM)
  • Structure-activity mapping: follow-up SPR work mapped the cyclization site via cysteine-to-serine mutations, and alanine scanning pinpointed five residues essential for binding in one of the two peptides
